چکیده
The entire central nervous system develops from the neural plate, initially a single-cell layered epithelium. Anterior–posterior patterning is established by signals emanating from tissues outside the neural plate, while local signaling centers that are established within the neural plate itself will refine this initial patterning. One signaling center that plays an important role in the early development of midbrain and anterior hindbrain is the mid/hindbrain organizer (MHB). It is expresses Wnt1 on its rostral side and Fgf8 on its caudal side to exert its effects on future midand hindbrain. We have previously shown that Sip1/Zfhx1b is a multi-domain transcriptional repressor that can interact with several protein partners, thereby exerting several different functions depending on cellular context. We report here a new function of Sip1 in midbrain and hindbrain patterning. At E8.5, Sip1 is expressed in almost the entire neural plate. Sip1 represses Dkk1 expression in the early midbrain, leading to precocious Dkk1 expression at late headfold stage and increased expression at 5S stage in Sip1 knockout embryos. As Dkk1 is an inhibitor of Wnt signaling, the observed aberrant expression of Dkk1 leads to disturbed Wnt–b-catenin signaling in the future midbrain and anterior hindbrain. Hence, this aberrant Wnt signaling in Sip1 knockout embryos leads to early MHB patterning defects, as shown by the altered expression domain of MHB region genes, including En2 and Pax5. Sip1 is thus a regulator of Wnt–b-catenin signaling during early midbrain and anterior hindbrain development and thereby indirectly influences patterning of these tissues.

The transcriptional responses of drug treatments combined with a protein target prediction algorithm was utilised to associate compounds to biological genomic space. This enabled us to predict efficacy of compounds in cMap and LINCS against 181 databases of diseases extracted from GEO. 18/30 of top drugs predicted for leukemia (e.g. Leflunomide and Etoposide) and breast cancer (e.g. Tamoxifen a...
متن کاملAnalysis of Promyelocytic Leukemia in Human Embryonic Carcinoma Stem Cells During Retinoic Acid-Induced Neural Differentiation
Background: Promyelocytic leukemia protein (PML) is a tumor suppressor protein that is involved in myeloid cell differentiation in response to retinoic acid (RA). In addition, RA acts as a natural morphogen in neural development. Objectives: This study aimed to examine PML gene expression in different stages of in vitro neural differentiation of NT2 cells, and to investigate the possible role o...
متن کاملI-13 Infertility with Impaired Zona Pellucida Adhesion of Spermatozoa from Mice LackingTauCstF-64
Background: Fertilization is a multistep process requiring spermatozoa with unique cellular structures and numerous germ cell-specific molecules that function in the various steps. In the highly coordinated process of male germ cell development, RNA splicing and polyadenylation help regulate gene expression to ensure formation of functional spermatozoa. Male germ cells express tauCstF-64 (Cstf2...
متن کاملADAM Gene Expression in The Adult CNS and Genetic Aberrations in Cancer Cells
ADAM metalloprotease-disintegrins share a common modular structure of functional domains for proteolytic, cell adhesion, and signaling interactions. The metalloprotease domain of oughly half of the known ADAMs contain an intact consensus metzincin catalytic site, and they are thus thought to function as active metalloproteases. The types of interactions mediated by ADAMs are expressly conspicu...

Purpose: The present study intends to show the characteristics of cultured limbal stem cell (CLSCs) and to compare them with normal Conjunctival (C), Limbal (L) and Cornea (K) tissues. Materials and Methods: The expressions of a set of genes potentially involved in differentiation and stemness function of limbal stem cells were assessed in freshly prepared limbal, corneal, and conjunctival tis...
